I'm backing up a group of VM's by selecting them by datastore, in example: The datastore of 6 TB hold 30 VM's, most of them are small,  1 is a large one taking 4TB of the space.

I would like when the backup starts that it creates a backup of that large VM first, reason being is that this one takes the most time to process, the other streams can then be used while the big server is being processed.

Is there a way to set priority?

You can set the priority on the client that is created for the VM and it will be honored by the VSA backup jobs.  However for the specific condition that you called out; there is already default priority logic that will prioritize a larger VM over a smaller VM.

Smaller VM's will be placed at higher priority over smaller, but not over newer VM's and those with more disks (https://documentation.commvault.com/commvault/v11/article?p=97216.htm).  Separating the VM into it's own subclient(VM Group) schedule is what you will need to do in order guarantee it starts when you want.
